The purpose of this study was to look into the anxiolytic effect of Punica granatum Linn. methanolic leaves extract in mouse experimental models. Punica granatum leaf methanolic (MLPG) extracts were evaluated for various phytochemical tests and pharmacological screening of MLPG such as hole board method light and dark box test motor in-coordination by Rota-rod apparatus elevated plus maze. The average time for a normal animal to fall off the Rota-rod apparatus is 3 to 5 minutes. Fall-off time was significantly reduced in group 4, MLPG (100mg/kg) due to motor in-coordination. The time spent in open arm entries increases with extract treated dose 2 (group 4). Diazepam treatment (group 2), extract treatment dose 1 (group 3), and extract treatment dose 2 (group 3) result in a significant reduction in time spent in close arm. The diazepam-treated group spends significantly more time in the lit box than the normal control group, whereas the MLPG-treated group spends significantly less time in the lit box than the diazepam-treated group. Diazepam altered several patterning’s, primarily those involving point-sniff and static-dip. Group 4's percent distribution and behavioral sniffing and dipping patterns have also changed significantly.
